Monoclonal antibodies to the human insulin receptor block insulin binding and inhibit insulin action.
Antibodies to the insulin receptor were prepared in BALB/c mice by immunization with IM-9 human lymphocytes, a cell type that has a large number of plasma membrane insulin receptors. The spleens of these mice were then removed, and their lymphocytes were fused to a mouse myeloma cell line, FO cells. After screening over 1,200 resulting hybrids, one stable hybrid was obtained that produced IgG1 ...
متن کاملInsulin receptor monoclonal antibodies that mimic insulin action without activating tyrosine kinase.
HTC rat hepatoma cells were transfected with human insulin receptor cDNA to a level of 40,000 receptors/cell. In these cells, as well as in nontransfected cells, insulin stimulated the uptake of alpha-aminoisobutyric acid. Two monoclonal antibodies directed against the human insulin receptor alpha subunit, like insulin, stimulated amino acid uptake in transfected HTC cells, but not in nontransf...

Monoclonal antibodies previously shown to react with five distinct epitopes on the human insulin receptor were tested for their metabolic effects on isolated human adipocytes. Two antibodies which reacted with receptor alpha-subunit and completely inhibited 125I-insulin binding mimicked the actions of insulin to stimulate lipogenesis from [14C]glucose and to inhibit catecholamine-induced lipoly...
متن کاملInsulin-binding antibodies in relation to insulin therapy.
Methods for detecting insulin binding or transporting antibodies are described, involving the use of radioactively labelled insulin (insulin (131)I). These antibodies are detected in a high proportion of patients receiving insulin, particularly those requiring large doses. The protein fractions concerned appear to be the beta and gamma globulins. The significance of the phenomenon is discussed.
متن کاملInsulin-mimetic anti-insulin receptor monoclonal antibodies stimulate receptor kinase activity in intact cells.
In the present studies, nine different monoclonal antibodies to the extracellular domain of the insulin receptor were tested in three different cell types for their ability to stimulate the intrinsic tyrosine kinase activity of the receptor. Previous studies had suggested that several of these monoclonal antibodies stimulate biological responses without stimulating the intrinsic tyrosine kinase...
